Abstract

PHB is produced by microorganisms (such as Ralstonia eutrophusMethylobacterium rhodesianum Or Bacillus megaterium) apparently in response to conditions of physiological stress, mainly conditions in which nutrients are limited.. In present study HM medium was used for the isolation of PHB producer halophiles, because it containe high salt concentration. Among them 21 isolates were found from the four different marine samples from various sites in Valsad district. Secondary screening has been done using M medium and the final screening was carried out with HM medium containing Nile blue A dye. After the final screening total 7 different isolates as PHB producers were selected and further studied. PHB production by selected isolated strain was carried out by Fed-batch fermentation in flask. Crude PHB extraction was carried out using ethyle acetate extract and further characterized by FTIR study.
