Abstract

The present study was aimed to identify the cellular mechanisms underlying lead induced hepatic injury by investigating the cell viability, antioxidant enzymes and transaminase enzymes using chang cell line ( human normal liver cell line) as model. Cell viability was determined by MTT assay.  Chang cells were cultured with different concentration of lead to find out the IC50 of lead. Based on the calculated concentration of lead, further experiments were conducted at two different time interval.  At 2nd hour and 6th hour, cellular levels of glutathione, superoxide dismutase (SOD), alanine aminotransferase (ALT), asparatate aminotransferase (AST), lactate dehydrogenase (LDH), Lipid peroxidation and Total protein were measured. Lead treatment demonstrated a time dependent increase in intracellular levels of reactive oxygen species (ROS) and decrease in superoxide dismutase (SOD) and reduced glutathione (GSH) levels. Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST), lactate dehydrogenase( LDH), Lipid peroxidation levels were increased in lead treated cells. The results conclude that one of the mechanism underlying lead induced hepatotoxicity could be due to oxidative stress.
