Abstract

Carissa carandas (L.) is an important medicinal plant and its roots, fruits, leaves are used for various health ailment treatments. Due to its excessive collection from natural habitat, destructive harvesting techniques, coupled with poor seed germination with less viability and low vegetative multiplication ratio and in order to contribute its production systems, an efficient protocol was developed for in vitro multiple shoot induction through node and shoot apex. During the studies adenine sulphate (Ads) has been used as an experimental hormone for the production of multiple shoots. Maximum numbers of shoots from nodal explant were produced on MS medium supplemented with 1.5 mg/l BAP + 1.0 mg/l Kn + 1.0 mg/l TDZ + 15 mg/l Ads. Same concentration i.e. 1.5 mg/l BAP + 1.0 mg/l Kn + 1.0 mg/l TDZ+ 15 mg/l Ads induced large number of shoots from shoot apex. In both explant cultures adenine sulphate played an important role for improving the frequency of multiple shoot induction when compared to culture media without adenine sulphate. Rooting was readily achieved upon transferring the shoots on to half- strength MS medium supplemented with 2.0 mg/l IAA. Micropropagated plantlets hardened in the green house were successfully established. This method could be effectively applied for the conservation and clonal propagation to meet the demand of planting materials.
