STUDIES ON THE PREPARATION OF A NOVEL SUPERMIX DNA LADDER BY RESTRICTION DIGESTION
Modern biological tools of recombinant technology have tremendous potential to characterize DNA molecules with the choice of DNA ladders. Determination of molecular weight of linear and double stranded DNA fragments either on agarose or polyacrylamide gel is done extensively with the DNA ladders. The most popular DNA ladders available in the market range from less than 25bp to 40 kb or more. Amalgamation of predetermined or definite size Polymerase Chain Reaction products to achieve novel DNA marker is the most common practice. In this study, we have described a method by which two novel supermix DNA ladders were prepared based on restriction digestion. The strategy was based on single, double and triple restriction digestion of plasmid and genomic DNA from various sources. The samples were run on 1% agarose gel, and the obtained pattern was analysed against available commercial ladders. The supermix DNA ladder constructed can be used in laboratory for analytical and diagnostic purposes in the field of molecular biology.
